Item Detección del hongo chytridiomycota (Batrachochytrium dendrobatidis) por técnica de PCR, en la rana marsupial andina (Gastrotheca riobambae) en la ciudad de Latacunga(2023-03) Jordán Bayas, Marjorie Vanessa; Avilés Esquivel, Diana FernandaIn the present research work, a total sampling area of approximately 7,200 m2 (T1 and T2) and 4,800 m2 (T3) was covered, where data were obtained on local distribution, preference for different types of substrate, population structure, as well as the presence of Batrachochytrium dendrobatidis. The main objective of this study was to determine the presence of Batrachochytrium dendrobatidis in the populations of adult frogs and tadpoles of Gastrotheca riobambae by endpoint PCR in the sectors of Belisario Quevedo (-0. 962806, -78.594406), Juan Montalvo (-0.900921, -78.605251), San Buenaventura (- 0.886293, -78.608960), belonging to the city of Latacunga, for this purpose a nonexperimental, transectional design was applied and for sampling a non-probabilistic method by convenience. For the sampling of adult frogs, standardized techniques for amphibians were applied, such as the survey method by visual encounters and fixed auditory points. The end-point PCR technique was applied to detect the presence of B. dendrobatidis and specific primers for the chytrid were used. Positive controls were used in each test performed, thus ensuring that the PCR conditions were adequate and amplified all the samples that were positive. The diagnostic assay was performed on 39 samples obtained by swabs from the three localities of Latacunga, the results obtained may be related to multiple factors such as sampling sites, ambient temperature or altitude. The three locations where samples were taken were sites with a certain degree of anthropic disturbance, which is one of the determining factors for the presence of B. dendrobatidis.
