Abstract: | Bacteria isolated from glacial, Andean and Antarctic ecosystems were identified. By rep-PCR using the BOX primer, 72 strains were genotypically characterized and a dendrogram was constructed by numerical taxonomy, using the Pearson coefficient and the UPGMA algorithm, obtaining the formation of 10 groups. From these, representatives were selected for the sequencing of the 16S rDNA gene and phenotypic characterization. The phylogenetic analysis of the 16S rDNA gene was performed by calculating the evolutionary distances of Jukes & Cantor and the Neighbor-Joining algorithm, allowing the identification of 11 different species. Of which, strains DG-222 and DG-203 probably belong to new species. However, further studies are necessary to demonstrate this. Through the phenotypic characterization, a dendrogram of numerical taxonomy of phenotypic data was constructed using the SSM coefficient and the UPGMA algorithm, obtaining the formation of 21 phenetic groups with 84,6 percent similarity. Most of the collection was determined as Gram negative, psychrotroph, neutrotolerant, weak halophilic or halophilic. In addition, new physiological characteristics of 4 different species were discovered. The analysis of polyphasic taxonomy with the strains that had fingerprinting patterns similar to the identified species, managed to locate 35 strains according to the molecular characterization. Forming 16 phenetic groups at 90 percent similarity and determining the polyphase taxonomic position of 48,6 percent of the collection. Probably, the diversity of the collection is underestimated and there could be species that have not been identified in this study. It is necessary to carry out a second analysis of the strains that were excluded in the analyzes. |
Description: | Se identificaron bacterias aisladas de ecosistemas glaciares, Andinos y Antárticos. Mediante rep-PCR usando el primer BOX, se logró caracterizar genotípicamente 72 cepas y se construyó un dendrograma por taxonomía numérica, usando el coeficiente de Pearson y el algoritmo de UPGMA, obteniendo la formación de 10 grupos. A partir de estos, fueron seleccionados representantes para el secuenciamiento del gen del ADNr 16S y caracterización fenotípica. El análisis filogenético del gen del ADNr 16S se realizó mediante el cálculo de las distancias evolutivas de Jukes & Cantor y el algoritmo de Neighbour-Joining, permitiendo identificar 11 especies distintas. De las cuales, las cepas DG-222 y DG-203 probablemente pertenezcan a especies nuevas. Sin embargo, es necesario realizar estudios posteriores para demostrar esto. Mediante la caracterización fenotípica se construyó un dendrograma de taxonomía numérica de datos fenotípicos utilizando el coeficiente SSM y el algoritmo UPGMA, obteniendo la formación de 21 grupos fenéticos al 84,6 por ciento de similitud. La mayor parte de la colección se determinó como Gram negativa, psicrótrofa, neutrotolerante, halófila débil o halófila. Además, se descubrió nuevas características fisiológicas de 4 especies distintas. El análisis de taxonomía polifásica con las cepas que tuvieron patrones de fingerprinting similares a las especies identificadas, logró ubicar 35 cepas en función a la caracterización molecular. Formando 16 grupos fenéticos al 90 por ciento de similaridad y determinando la posición taxonómica polifásica del 48,6 por ciento de la colección. Probablemente, la diversidad de la colección se encuentra subestimada y podrían existir especies que no se han identificado en este estudio. Es necesario realizar un segundo análisis de las cepas que fueron excluidas en los análisis. |
