Abstract

Polyethylene terephthalate PET accumulates in ecosystems at rates that conventional recycling methods cannot compensate for. The PETase from Ideonella sakaiensis, IsPETase, degrades PET, but its thermostability below 40 degrees Celsius limits its use in industrial applications. The structural comparison with the thermophilic cutinase LCC, whose D193 residue participates in salt bridge networks that sustain activity up to 70 degrees Celsius, motivated the S188D variation, which replaces the TCC codons with GAC, based on the ancestral mutant M8 architecture of IsPETase. The variant was cloned into the pET-30a(+) vector and expressed in E. coli Rosetta (DE3) under induction with IPTG 0.1 mM at 20 degrees Celsius. Purification by IMAC Ni NTA affinity chromatography produced 5,662 g of soluble protein, with fractions four and five showing the highest active protein concentration, 2,49 mg per mL, and the presence of the protein between the 26 to 34 kDa band was confirmed by SDS PAGE. The degradation assay showed that the activity of 1.5 mg of enzyme per g of micronized post consumer PET, without prior amorphization, at 50 degrees Celsius for 72 hours, reduced PET by an unspecified percentage compared with the control sample. This performance is acceptable, but it does not show significance compared with ancestral mutant variants. Therefore, it is necessary to integrate S188D with other cumulative mutations that strengthen the functional stability of IsPETase and consolidate it as a viable strategy for plastic biodegradation at an industrial scale.

El polietileno tereftalato (PET) se acumula en ecosistemas a tasas que los métodos convencionales de reciclaje no logran compensar. La PETasa de Ideonella sakaiensis (IsPETasa) degrada el PET, pero su termoestabilidad por debajo de 40 grados centígrados limitan su aplicación industrial. La comparación estructural con la cutinasa termofílica LCC, cuyo residuo D193 participa en redes de puentes salinos que sostienen la actividad hasta 70 grados centígrados, motivó la variación S188D que sustituye los codones TCC por GAC, sobre la arquitectura mutante antecesora M8 de IsPETasa. La variante se clonó en el vector pET-30a(+) y se expresó en E. coli Rosetta (DE3) bajo inducción con IPTG 0,1 mM a 20 grados centígrados. La purificación por cromatografía de afinidad IMAC-Ni-NTA produjo 5,662 g de proteína soluble, registrando a las fracciones 4 y 5 con mayor concentración proteica activa (2,49 mg por mL), y se confirmó, por SDS-PAGE, la presencia de la proteína entre la banda de 26 a 34 kDa. El ensayo de degradación mostró que la actividad de 1,5 mg enzima g PET post-consumo micronizado y sin amorfización previa, a 50 grados centígrados durante 72 horas, redujo el PET un 13,22 por ciento a comparación de la muestra control. Este rendimiento es aceptable pero no tiene significancia a comparación de variantes mutantes antecesoras, por lo que es necesario integrar a S188D con otras mutaciones acumulativas que refuercen la estabilidad funcional de IsPETasa y la consoliden como una estrategia viable para la biodegradación del plástico a escala industrial.
