Influencia de fitohormonas en la regeneración in vitro de Solanum tuberosum L. vía organogénesis directa
(2023-03) Aleaga Villacís, Pedro Iván; Leiva Mora, Michel
The present investigation was developed with the purpose of determining how different phytohormones influence in vitro regeneration of Solanum tuberosum L. Var. Superchola via direct organogenesis, among which were included concentration and type of growth regulator. In the in vitro establishment, the type of auxins and their concentration were analyzed. In the in vitro multiplication phase, the concentration and type of cytokinins were analyzed. In the in vitro rooting phase, the effect of auxin type and concentrations were determined. In the acclimatization of the plants, the type of substrate was analyzed. A completely randomized design was used in each experiment. Nodal segments of potato Var. Superchola were established without the addition of auxins (Control), however, the plants presented a higher percentage of oxidation. In the in vitro multiplication phase of nodal segments, concentrations of 3 mg.L-1 of 2iP, 1 mg.L-1 of 6-BAP and 1.5 mg.L-1 of KINETIN increased the number of shoots, number and length of internodes, number of stems and plant height. In the in vitro rooting phase, concentrations of 0.5 mg.L-1 of AIA, 0.5 mg.L-1 of ANA and 0.2 mg.L-1 of AIB increased the rooting percentage, number and length of roots, stem thickness and number of leaves per plant. In the acclimatization phase, the plants adapted regardless of the type or dosage of substrate. With the results of the present work it was possible to obtain acclimatized plants of Solanum tuberosum L. of the Superchola variety capable of developing under greenhouse conditions.
