Abstract: Breast cancer is the second leading cause of death in women around the world, despite advances in medicines and therapies, it remains a public health problem, for this reason it is still necessary to investigate new active compounds that could become candidates for the development of new drugs. In this context, Ecuador is a highly diversified country with a significant number of ancestral plants to which indigenous communities attribute anti-proliferative and anti-carcinogenic properties. The objective of the research was to evaluate the effect on the expression of the Bax and bcl-2 genes by polymerase chain reaction in real time (qrt-PCR) extracts of Ilex guayusa, Uncaria tomentosa and Croton lechleri in the MCF7 breast cancer cell line. The extracts were obtained by maceration in ethanol at 96%, lyophilization and were resuspended in PBS 1X. The MFC-7 cell line was treated with the various extracts at a concentration of 3ug/ul for 24 hours. The total RNA was then extracted from the treated cells and its concentration was determined. cDNA was then synthesized and the expression of the Bax and bcl-2 genes was evaluated using a real time PCR. The obtained results showed that the ethanolic extract of Croton lechleri increased the expression of the Bax gene (p<0.05) and decreased the expression of bcl-2 (p<0.001) in relation to MCF7 cells not treated with the extract. The other two excerpts were inconclusive. The results were analyzed using variance analysis (ANOVA) in addition to a post-hoc Tukey analysis. The significance for Bax was <0.0002 and <0.0001 for bcl-2 between treatments, which is why the alternate hypothesis was accepted.
Description: El cáncer de mama es la segunda causa de muerte en mujeres alrededor del mundo, a pesar de los avances en medicamentos y terapias, sigue siendo un problema de salud pública, por este motivo todavía es necesario investigar sobre nuevos compuestos activos que podrían constituirse en candidatos para el desarrollo de nuevos fármacos. En este contexto, el Ecuador es un país megadiverso que cuenta con una cantidad significativa de plantas de uso ancestral a las cuales, las comunidades indígenas les atribuyen propiedades anti-proliferativas y anti-carcinogénicas. El objetivo de la investigación fue evaluar el efecto sobre la expresión de los genes Bax y Bcl-2 por reacción en cadena de la polimerasa en tiempo real (qRT-PCR) de los extractos de Ilex guayusa, Uncaria tomentosa y Croton lechleri en la línea celular de cáncer de mama MCF7. Los extractos se obtuvieron mediante maceración en etanol al 96%, liofilización y fueron resuspendidos en PBS 1X. La línea celular MFC-7 fueron tratados con los diversos extractos a una concentración de 3ug/uL durante 24 horas. A continuación, se extrajo el RNA total de las células tratadas y se determinó su concentración. Entonces se sintetizó el cDNA y se evaluó la expresión de los genes Bax y Bcl-2 mediante una PCR a tiempo real. Los resultados obtenidos mostraron que el extracto etanólico de Croton lechleri aumentó la expresión del gen Bax (p<0.05) y disminuyó la expresión de Bcl-2 (p<0.001) con relación a las células MCF7 no tratadas con el extracto. Los otros dos extractos fueron no concluyentes. Los resultados fueron analizados mediante el análisis de varianza (ANOVA) además de un análisis post-hoc Tukey. La significancia para Bax fue <0.0002 y <0.0001 para Bcl-2 entre los tratamientos, razón por la cual se aceptó la hipótesis alterna.
