Título : Efecto del aceite de canela (Cinnamomum zeylanicum Blume) en Botrytis cinerea Pers. IN VITRO e IN VIVO

Resumen : To evaluate the effect of cinnamon oil (Cinnamomum zeylanicum Blume) on Botrytis cinerea Pers. In vitro and in vivo. In the BIONIKA laboratories, located in the city of Quito. The factor under study was the application doses of the commercial product CINAMON, for this purpose, dilutions of 0.2, 0.3, 0.4, 0.5, 0.6, 0.7, 0.8, 0.9, 1.0 were prepared, 1,2, 1.3, 1.4 ml L-1. The concentration of the phytopathogen was log 6, both in the in vivo and in vitro tests, in addition an absolute control (without application of cinnamon oil) and a positive control with the application of a commercial fungicide (Ciprodinil + Fludioxonil) The treatments investigated were 14, both in vitro and in vivo. Statistically, a completely randomized block design (DBCA) with 4 replications was applied, analysis of variance and Tukey significant tests were performed at 1%, for the treatments that showed statistical significance. For the isolation of B. cinerea, the methodology developed in the Bionika laboratory was applied and involved the following phases: extraction, identification of the fungus and purification. The response variables were: the germination of conidia of Botrytis cinerea Pers., The evaluation process was started after 12 hours of inoculation, the data collection was made every 2 hours; for the corresponding calculations, the formula proposed by Cañedo & Ames was applied, and modified by Falconí: % GC = (number of germinated conidia / total number of conidia) * 100; the germinative tube growth rate (vctg) was measured at intervals of 4 hours, prior to the 24-hour inoculation, and the formula proposed by Arango et. Al. And modified by Falconi: vctg (mm / d) = germ tube growth (mm) / hours of incubation; the modes of action were determined organographically by means of Cytohistochemistry. The evaluation of the effect of cinnamon oil on the phytopathogen determined that the best dose was 1.4 ml x L-1 since it inhibited the growth of the fungus and its proliferation in vitro and in vivo. In the establishment to determine the oil dose, a previous test was carried out with doses of 0.25 ml x L-1, 0.50 ml x L-1 and 1.0 ml x L-1. The results of the in vivo test were analyzed organographically the potential sites of action, and their effect by the active ingredients of the cinnamon oil CINNAMON ©. In this case the effects were evidenced from the dose 1.0 ml x L-1 up to 1.4 ml x L-1.
Descripción : Con el propósito de evaluar el efecto del aceite de canela (Cinnamomun zeylanicum Blume) sobre Botrytis cinerea Pers. in vitro e in vivo. En en los laboratorios BIONIKA, ubicado en la ciudad de Quito. El factor en estudio fue las dosis de aplicación del producto comercial CINAMON, con este propósito, se prepararon diluciones de 0.2, 0.3, 0.4, 0,5, 0,6, 0,7, 0,8, 0,9, 1.0, 1,2, 1.3, 1.4 ml L-1. La concentración del fitopatógeno fue de log 6, tanto en los ensayos en in vivo como en in vitro, además se empleó un testigo absoluto (sin aplicación de aceite de canela) y de un testigo positivo con la aplicación de un funguicida cuyos ingredientes activos son Ciprodinil+Fludioxonil. Los tratamientos investigados fueron 14, tanto in vitro como in vivo. Estadísticamente se aplicó un diseño de bloques completamente al azar (DBCA) con 4 repeticiones, se efectuaron análisis de varianza y pruebas significativas Tukey al 1%, para los tratamientos que demostraron significación estadística. Para el aislamiento de B. cinerea, se aplicó la metodología desarrollada en el laboratorio Bionika y que implica las siguientes fases: extracción, identificación del hongo y purificación. Las variables respuesta fueron: la germinación de conidias de Botrytis cinerea Pers., El proceso de evaluación se inició luego de 12 horas de inoculación, la toma de datos se efectuó cada 2 horas; para los cálculos correspondientes se aplicó la fórmula propuesta por Cañedo y Ames, y modificada por Falconí: % GC=(número de conidias germinadas/número total de conidias)* 100; la velocidad de crecimiento del tubo germinativo (vctg), se midió a intervalos de 4 horas, previo a la inoculación de 24 horas y se aplicó la fórmula propuesta por Arango et. Al. y modificado por Falconi: vctg (mm/d) = crecimiento tubo germinativo (mm)/ horas de incubación; los modos de acción se determinaron organográficamente por medio de Cito Histoquímica. La evaluación del efecto del aceite de canela en el fitopatógeno determinó que la mejor dosis fue de 1.4 ml x L-1 ya que inhibió el crecimiento del hongo y su proliferación en in vitro e in vivo. En el establecimiento para determinar la dosis de aceite se realizó un previo ensayo con dosis de 0.25 ml x L-1, 0.50 ml x L-1 y 1.0 ml x L-1. Los resultados del test in vivo, se analizaron organográficamente los potenciales sitios de acción, y su efecto por los ingredientes activos del aceite de canela CINAMON©. En
